With more than 170,000 publications in 2016, inflammation remains a “hot” topic in today’s Life Science research (source: Google Scholar). Interestingly, the ELISA test is still one of the most popular tools for the accurate quantification of human cytokines involved in this biological process. Here, we review the 5 protein targets most studied by our clients, together with their favourite ELISA kits by RayBiotech in the field of inflammation.

Target # 1 – Human Interleukin-6 (IL-6)

IL-6 is active in inflammation (either acute or chronic), and in B cell maturation. This cytokine is implicated in a wide variety of inflammation-associated disease states (incl. diabetes mellitus, rheumatoid arthritis…). To quantify human IL-6 in cell culture supernatants, plasma, and serum samples, researchers use RayBiotech‘s Human IL-6 Sandwich-based ELISA kit (cat. nr ELH-IL6-1).

This RayBio® Human IL-6 ELISA Kit has a sensitivity of 3 pg/ml (detection range: 3 pg/ml – 1,000 pg/ml) with no cross-reactivity with any of the cytokines tested: Human Angiogenin, BDNF, BLC, ENA-78, FGF-4, IL-1 alpha, IL-1 beta, IL-2, IL-3, IL-4, IL-5, IL-7, IL-8, IL-9, IL-10, IL-11, IL-12 p70, IL-12 p40, IL-13, IL-15, I-309, IP-10, G-CSF, GM-CSF, IFN-gamma, Leptin (OB), MCP-1, MCP-2, MCP-3, MDC, MIP-1 alpha, MIP-1 beta, MIP-1 delta, PARC, PDGF, RANTES, SCF, TARC, TGF-beta, TIMP-1, TIMP-2, TNF-alpha, TNF-beta, TPO, VEGF.

For Lysates, it is recommended to use the RayBio® Human IL-6 ELISA Kit for cell and tissue lysate samples (cat. nr ELH-IL6-CL-1).

Target # 2 – Human TNF-alpha

TNF-alpha is a proinflammatory cytokine that regulates multiple biological processes (incl. cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation). TNF is implicated in a variety of diseases such as autoimmune diseases, insulin resistance, and cancer.

The RayBio® Human TNF alpha ELISA Kit (cat. nr ELH-TNFa-1) is for cell culture supernatants, plasma, and serum samples with a sensitivity of 30 pg/ml with a detection range between 30 pg/ml and 6000 pg/ml). For Lysates, you will likely prefer the format dedicated for such biological samples: cat. nr ELH-TNFa-CL-1.

The Human TNF-a ELISA kit shows no cross-reactivity with any of the cytokines tested: Human ANG, CD23, Eotaxin, GCSF, GM-CSF, GRO-alpha, GRO-beta, GRO-gamma, I-309, IFN-gamma, IL-1 alpha, IL-1 beta, IL-3, IL-4, IL-5, IL-6, IL-7, IL-8, IL-10, IL-12 (p40), IL-12 (p70), IL-15, IL-16, IP-10, MCP-1, MCP-2, MCP-3, MCP-4, MCSF, MIG, MIP-1 alpha, MIP-1 beta, NAP-2, PDGF, PF-4.

Target # 3 – IL1-beta

IL1-BETA is an important mediator of inflammatory response and is involved in a variety of cellular activities (incl. cell proliferation, differentiation, and apoptosis). The induction of cyclooxygenase-2 (PTGS2/COX2) by this cytokine in the CNS contributes to inflammatory pain hypersensitivity. This cytokine has also been described to be active in cancer / immune cell crosstalk a.k.a. immuno-editing.raybio-elisa-kit-by-raybiotech-and-tebu-bio

The RayBio® Human IL-1 beta (IL-1 F2) ELISA Kit is for cell culture supernatants, plasma, and serum samples with a sensitivity of 0.3 pg/ml (detection range: 0.3 pg/ml – 100 pg/ml; cat. nr ELH-IL1b-1).

This ELISA kit shows no cross-reactivity with any of the cytokines tested: Human Angiogenin, BDNF, BLC, ENA-78, FGF-4, IL-1 alpha, IL-2, IL-3, IL-4, IL-5, IL-7, IL-8, IL-9, IL-11, IL-12 p70, IL-12 p40, IL-13, IL-15, I-309, IP-10, G-CSF, GM-CSF, IFN-gamma, Leptin (OB), MCP-1, MCP-2, MCP-3, MDC, MIP-1 alpha, MIP-1 beta, MIP-1 delta, PARC, PDGF, RANTES, SCF, TARC, TGF-beta, TIMP-1, TIMP-2, TNF-alpha, TNF-beta, TPO, VEGF.

Target # 4 – IL-1 R4 / ST2 (IL-33 receptor)

IL-33 receptor or ST2 (IL-1 R4) when bound to IL-33 elicits either pro- or anti-inflammatory effects depending on the disease and the model. IL-33 is also considered to be a promising cardiovascular biomarker.

The RayBio® Human IL-1 R4 / ST2 (IL-33 receptor) ELISA Kit is for cell culture supernatants, plasma, and serum samples (2 pg/ml as sensibility threshold and with 2 pg/ml – 1,200 pg/ml as a detection range – cat. nr ELH-IL1R4-1).
