1 – Sircol™ Soluble Collagen Assay

The Sircol™ Collagen Assay is a dye-binding method for the analysis of acid and pepsin-soluble collagens. The Sircol Assay is suitable for monitoring collagen produced in situ or during in vitro cell culture and in vitro extracellular matrix, (ECM), formation:

in vivo

– collagen, soluble in cold acid or pepsin, recovered from mammalian soft tissues, cartilages and fluids.

in vitro

– collagen, soluble in cold acid or pepsin, released into cell culture medium during cell growth and cell maintenance.
– collagen, soluble in cold acid or pepsin, recovered from newly formed extracellular matrix that has been deposited onto cell culture treated plastic surfaces, (T-flasks and microwell plates).

This easy-to-use colorimetric assay, will allow you to detect up to 1µg of soluble Collagen in less than two hours. Types I to V mammalian collagens can be measured with the assays.

2 – Sircol™ Insoluble Collagen Assay

The Sircol™ INSOLUBLE Collagen Assay is a dye binding method for the analysis of insoluble collagen fibers.

The assay can be used to assess the production rate of newly laid down collagen fibers during periods of rapid growth, development, tissue repair, remodelling and wound healing. Sources of material includes tissues, bone and calcified tissue.

  •  It replaces the conventional hydroxyproline analysis. No more working with concentrated acids or alkalis!
  • The Sircol Insoluble Assay uses mild acid and temperature treatment for 2 to 3 hrs.
  • The solubility conversion to denatured collagen and subsequent measurement can be completed within one working day.
  • The ratio of soluble and insoluble collagens can be measured using the same Sircol Dye Reagent.

Of course, by adding the results together a value for the “Total Collagen” in a sample can be established.

3 – Blyscan™ Glycosaminoglycan Assay

The Blyscan™ Assay is a quantitative dye-binding method for the analysis of sulfated proteoglycans and glycosaminoglycans (sGAG) in one hour. Test material can be assayed directly from:

Soluble extract
  • Solid tumors – Arteries – Heart – Skin and all materials containing ECM
  • Fibrous and Hyaline Cartilages
  • sGAG from Synovial fluid, Urine and Gel Chromatography fraction Aliquots

With this colorimetric assay, an extensive range of Mammalian Proteglycans can be measured:

  • Chondritin Sulfates / Keratan Sulfates / Dermatan Sulfates / Heparan Sulfates (Including Heparin)

Used to measure up to 0,25µg of total sGAG, this Blyscan™ Glycosaminoglycan Assay can be also adopted to determine the O– and N-sulfated glycosaminoglycan ratio within test samples.

4 – Fastin™ Elastin Assay

The Fastin™ Elastin Assay is a quantitative dye-binding method (Read out: 513 nm) for the analysis of elastins extracted from mammalian sources (Tissue extracts and cells during in-vitro culture).


Elastin forms that can be measured by the Fastin Assay as α-elastin:

  • soluble tropoelastins
  • lathyrogenic elastins
  • insoluble elastins (following solubilization to elastin polypeptides [α-elastin, κ-elastin])

The Purple-Jelley Hyaluronan Assay is used to measure the quantity of hyaluronan (hyaluronic acid) in mammalian tissue. As for the previous assay, this assay is a dye-based method with a detection limit of 0.2μg.

This assay relies on the affinity of the Purple dye to Hyaluronic acid. To avoid cross reactivity of the dye, it’s essential to remove other potential binding protein (Tissues proteins and Sulfated glycosaminoglycans (sGAG)) before the measurement.

Methods for easy protein and sGAG removal are specified in the Purple-Jelley™ assay manual. Finally the readout of the assay can be done at 655 nm using a microplate spectrophotometer.

Distribution results of Hyaluronic Acid in different kind of tissues, summarized in this table, were all obtained with this Purple-Jelley Hyaluronan Assay.
