Tetramethylbenzidine (TMB) is a classic substrate for peroxidase activity that is used for microwell plate analysis such as an ELISA test. A quantitative color change from yellow to blue reveals the activity and can be read at 620-650 nm for ELISA test.
Substrate selection in immunoassay development
Surmodics have developed several types of TMB, for different purposes.
TMBW is the gold standard with fast reaction.
TMBS shows super high sensitivity (not shown on the previous image).
TMSK is the slow kinetics, TTMB is the super slow kinetics and TMBX offers an extended range.
For membranes instead of microwell plates, there are dedicated types of TMB. They can be very useful for Western blot without the need for a dark room. They are the standard TMBM and the enhanced version ESPM to catch low signals.
