JAK-STAT Pathway

The Janus kinase (JAK) / Signal Transducer and Activator of Transcription (STAT) pathway a key regulator of cellular proliferation, differentiation, migration, apoptosis and survival. Numerous cytokines, growth factors and Interferons interact with the JAK/STAT signaling pathway which in turn   regulates many biological processes, especially in hematopoietic development and immune responses.

The signaling is initiated upon the binding of cytokines (like Cardiotrophin-1, IFNs, LIF, TGF, EGF, bFGF…) to their cognate receptors causing receptor dimerization  and trans-phosphorylation of the recruited JAKs. Once activated, JAKs phophorylate the STAT family members. These p-STATs in turn translocate to the nucleus and act as dimers to regulate the expression of target genes acting as transcription factors.

Dysregulation of JAK-STAT signaling leads to several immune disorders, such as hyperlgE syndrome, erythremia, and leukemia.
