PD-1 / NFAT Reporter – Jurkat Cell Line
The PD-1 reporter cell line is a T cell line expressing luciferase under the control of NFAT response elements. The level of the luciferase activity measured with the One-Step luciferase detection system (BPS Bioscience) corresponds to the T cell activation in response to the TCR activator from the target cell.
To escape, the target cell may present PD-L1 or PD-L2 to PD-1. Indeed, the interaction inhibits the T cell activation. It leads to decrease in luciferase activity.
The inhibition of PD-1 interaction to PD-L1/PD-L2 blocks the escape of the target cell and promotes the T cell activation. It can be performed by a specific neutralizing antibodies such as the Anti-PD-1 Neutralizing Antibody and the Anti-PD-L1 (CD274) Neutralizing Antibody. The inhibition of the PD-1/PD-L1 interaction leads to the rescue of the luciferase activity coded by the NFAT-Luc reporter (Similarly shown by fold induction).
The PD-1 reporter line is the key of a cell-based assay to investigate the escape of target cell, namely cancer cells, to be killed by the T cell.

Designing the target cell
The new TCR Activator / PD-L1 Mammalian Expression Kit allows you to transfect another cell line to express the TCR activator and the PD-L1 protein. These cells can be used together with the PD-1 reporter cell line for a cellular assay of PD-1:PD-L1 binding and activation. There is also a solution for PD-L2, the TCR Activator / PD-L2 Mammalian Expression Kit.
