Nerve growth factor(NGF) is a polypeptide involved in the regulation of growth and differentiation of sympathetic and certain sensory neurons. NGF is thought to have a profound effect on the development and maintenance of sympathetic and embryonic sensory neurones. NGF activity isolated from the male mouse submaxillary gland(MSG) consists of three types of subunits, alpha, beta and gamma, which specifically interact to form a 7S, approximately 130,000-molecular weight(Mr) complex. The 7S complex contains two identical 118-amino acid beta-chains, which are solely responsible for the nerve growth-stimulating activity of NGF. NGF, which is expressed by inflammatory cells and effects changes that lead to increased neural responsiveness, could be a pivotal mediator in allergic rhinitis.

Neurotrophin-4(NT-4), also known as neurotrophin-5(NT-5), is a protein that in humans is encoded by the NTF4 gene. Human NT-4 as well as a human NT-4 pseudogene colocalize to chromosome 19 band q13.3. NT-4 is a member of a family of neurotrophic factors, the neurotrophins, that control survival and differentiation of vertebrate neurons(2-4). NT-4 is a neurotrophic factor that signals predominantly through the TrkB receptor tyrosine kinase. Treatment with recombinant OSM leads to the inhibition of proliferation and changes in cellular morphology of a number of tumor cell lines derived from a wide variety of tissue types. OSM signals through cell surface receptors that contain the protein gp130. The type I receptor is composed of gp130 and LIFR, the type II receptor is composed of gp130 and OSMR.

Osteoprotegerin(OPG) is identical to osteoclastogenesis inhibitory factor(OCIF), a soluble member of the tumor-necrosis factor receptor family that inhibits osteoclastogenesis. OPG is considered to play an important role in the regulation of bone resorption by modifying osteoclast differentiation. Osteoprotegerin is an independent risk factor for the progression of atherosclerosis and onset of cardiovascular disease. It can act as a soluble factor in the regulation of bone mass and imply a utility for OPG in the treatment of osteoporosis associated with increased osteoclast activity. OPG system may play a critical role in the development of osteolytic bone disease in multiple myeloma and that targeting this system may have therapeutic potential.
