Alkaline phosphatase hydrolyzes the BCIP to 5-bromo-4-chloro-3-indole, which is oxidized by oxygen in the atmosphere, yielding a blue dye. In the presence of NBT, an insoluble dark blue precipitate is formed after a reduction reaction instead.
