DNA was prepared from cell line WM3061. WM3061 is a metastatic human melanoma cell line. These are small spherical cells that float easily after splitting and require 5% medium to adhere to culture flask. WM3061 cells produce xenograft tumors when injected into immunocompromised mice. This cell line contains a G13R mutation at position 13 in the N-RAS gene. Mutations affecting codon 13 lead to constitutive activation of RAS GTPase and ultimately neoplastic growth. The G13R is a rare NRAS mutation found in melanoma.

DNA was prepared from cell line WM3211. WM3211 is a tumorigenic (VGP) primary melanoma cell line with competence for metastasis. These cells display fibroblastic morphology in culture. This cell line was established from a metastatic site (lymph node) in a 34-year-old female. This cell line contains a L576P mutation at position 576 in the c-KIT gene. The L576P mutation results in an amino acid substitution at position 576 in KIT, from a Leucine (L) to a proline (P). This mutation occurs within the juxtamembrane domain. Mutant KIT proteins have increased kinase activity and transforming activity in vitro. This cell line is wild type for BRAF, PTEN, N-RAS, and CDK4. WM3211 cells produce xenograft tumors when injected into immunocompromised mice.

DNA was prepared from cell line WM3246. WM3246 is a metastatic human melanoma cell line that was established from an adrenal gland metastatic site of a female patient. This cell line expresses mutated PTEN gene and is wild type for BRAF, N-RAS, c-KIT and CDK4. WM3246 cells produce xenograft tumors when injected into immunocompromised mice.

DNA was prepared from cell line WM3248. WM3248 is a human melanoma cell line. This cell line features the specific V600E (Val600Glu) mutation at codon 600 in the BRAF gene. This mutation causes constitutively active kinase activity and activation of MEK and ERK signaling pathway. This cell line also expresses PTEN loss of function including mutated and hemizygous PTEN deletion and is wild type for N-RAS, c-KIT, and CDK4.
