SiR-azide is the 3-azidopropyl derivative of the fluorophore silicon rhodamine (SiR). Sir-azide reacts with terminal alkyne derivatives in the presence of copper (I) to form 1,2,3-triazoles linkage. It can also react with strained alkynes such as DIFO or DIBO without the need of copper. Small molecules, peptides, oligonucleotides or proteins bearing the appropriate alkyne can be conjugated to SiR-azide. SiR-azide is compatible with GFP and/or m-cherry fluorescent proteins. It can be imaged with a standard Cy5 filterset.

SiR-alkyne is the N-propargylcarboxamide derivative of the fluorophore silicon rhodamine (SiR). Sir-alkyne reacts with azide derivatives in the presence of copper (I) to form 1,2,3-triazoles linkage. Small molecules, peptides, oligonucleotides or proteins bearing the appropriate azide can be conjugated to SiR-alkyne. SiR excitation and emission are compatible with GFP and/or m-cherry fluorescent proteins. It can be imaged with a standard Cy5 filterset.

SiR-maleimide is the 2-maleimidoethyl derivative of the fluorophore silicon rhodamine (SiR). SiR-maleimide reacts with thiols to form covalent conjugates. Small molecules, peptides, oligonucleotides or proteins bearing a free thiol can be conjugated to SiR-maleimide. SiR excitation and emission are compatible with GFP and/or m-cherry fluorescent proteins. It can be imaged with a standard Cy5 filterset.

SiR-lysosome is based on the cathepsin D binding natural product pepstatin A. SiR-lysosome is fluorogenic, cell permeable and highly specific for lysosomes. Sir-lysosome stains lysosomes in ilve cells without the need for genetic manipulation or overexpression. Its emission in the far red minimizes phototoxicity and sample autofluorescence. SiR-lysosome is compatible with GFP and/or m-cherry fluorescent proteins. It can be imaged with standard Cy5 filtersets. SiR-lysosome can be used for widefield, confocal, SIM or STED imaging in living cells and tissue.

SiR-BCN is a bicyclo[6.1.0]nonyne (BCN) derivative of the fluorophore silicon rhodamine (SiR). SiR-BCN contains a short PEG linker between the SiR fluorophore and the BCN moiety. SiR-BCN reacts quickly with tetrazine and azide derivatives to form covalent linkages. Small molecules, peptides, oligonucleotides or proteins bearing the appropriate tetrazine or azide can be conjugated to SiR-BCN. SiR excitation and emission are compatible with GFP and/or m-cherry fluorescent proteins. It can be imaged with a standard Cy5 filterset.
