Boc-O-SN-38 is a derivative of SN-38, an anti-tumor payload that is often linked to ADCs to be directly delivered to cells. Its mechanism of action is inhibition of DNA topoisomerase I, halting DNA synthesis and causing single-strand breaks in DNA. The Boc protection allows for controlled reactions without affecting the amine functionality.
NH-bis(PEG8-t-butyl ester) is a bifunctional PEG linker with a pair of t-butyl esters joined together at a secondary amine. The secondary amine can be used as a nucleophile while the t-butyl esters can be deprotected to allow for reactions with nucleophiles such as amines or alcohols. The use of a PEG spacer increases aqueous solubility and will impact the ADME of the final PEG conjugate.
