SPY505-CA is the ChloroAlkane (CA) derivative of our SPY505 fluorophore. It emits light in the green part of the UV-ViS spectrum, it is fluorogenic, highly cell permeable and well suited for STED and SIM superresolution imaging. SPY505-CA can be imaged with a standard FITC or YFP filterset. It can be used for widefield, confocal, SIM or STED imaging in living or fixed cells and tissue. It has the same structure as MaP510-halo, the name used in its original publication in Nature Chemistry. ChloroAlkane (CA) is the substrate of the self labeling tag Halo-tag™*. Upon reaction with a CA derivative, Halo-tag™* forms a covalent bond with the substrate. Its optimized structure allows quick labeling of the golgi apparatus in live cells with high specificity and low background. SPY555-golgi stains the golgi apparatus in live cells without the need for genetic manipulation or overexpression of fluorescent proteins. Its absorbance and emission spectra are similar to TMR. SPY555-golgi enables multicolor imaging with SPY505, SPY595, SPY650, SPY700, SiR and GFP. SPY555-golgi can be imaged with standard TMR or Cy3 filtersets. CA_TestKit combines our 3 Halo-tag™ subtrates SPY505-CA, SPY555-CA and SiR-CA, offering a wide flexibility for Halo-tag™ fusion proteins labeling in live and fixed cells. The ligands are all bright, highly fluorogenic, and cell permeable. The 3 fluorophores are covering 3 distinct and very common imaging channels: FITC, TMR/Cy3 and Cy5.
