Production of eGFP mRNA Labeled with Fluorophore for Tracking of Delivery and Expression
We produced fluorescently labeled eGFP mRNA to help assess LNP encapsulation efficiency and monitor mRNA delivery in mammalian cells.
Using Andy Fluor-UTP, we generated fluorescent mRNA detectable at multiple wavelengths and confirmed its successful transfection and expression in HEK293 cells.

Can you provide mRNA coding for reporter genes?
Yes, we offer several off-the-shelf mRNAs for common reporter genes, including eGFP, mCherry, FLuc, GLuc. Other off-the-shelf mRNA proposed are OVA, and β-gal.

I have an amino acid sequence for a protein of interest. Can you produce the corresponding mRNA?
Absolutely. We can design an mRNA sequence based on your protein’s amino acid sequence. We also perform codon optimization to minimize immunogenicity and reduce the risk of truncated mRNA forms.

I already have a plasmid containing my sequence of interest. Can you produce mRNA from it?
Yes, we can synthesize mRNA directly from your plasmid, provided it is compatible with in vitro transcription. If not, we can insert your sequence into one of our optimized plasmids, featuring enhanced UTRs and polyA tails for efficient transcription and translation in mammalian cells.

What are the minimum and maximum gene sizes for in vitro transcription (IVT)?
For IVT, the minimum gene size is 100 base pairs. While there is no strict upper limit, transcripts over 6 kb are more complex to produce and may result in lower yield consistency under standard conditions. Our high-resolution quality control system is also more effective for smaller transcripts.

What quality control (QC) measures do you perform on LNPs?
We conduct Dynamic Light Scattering (DLS) to monitor LNP size and homogeneity (polydispersity index). We also assess encapsulation efficiency using mRNA quantification via Ribogreen Assay.

What is the shelf-life of LNPs?
LNPs are stable for up to 3 weeks at 4°C. For extended storage, the addition of a cryoprotectant (sucrose or trehalose) enables LNPs to be kept at -20°C for several months.

What transfection agents do you use for in vitro mRNA delivery?
For in vitro applications, we recommend using mRNA-fect or LNP formulations. mRNA-fect typically offers higher efficiency for in vitro delivery.
