Functional Assays

The METTL3/METTL14 Complex Chemiluminescent Assay Kit is designed to measure METTL3/METTL14 complex activity for screening and profiling applications. The key to the METTL3/METTL14 Complex Chemiluminescent Activity Assay Kit is a highly specific antibody that recognizes N6-methylated adenosine. With this kit, only three simple steps are required for methyltransferase detection. First, S-adenosylmethionine is incubated with a sample containing assay buffer and methyltransferase enzyme. Next, primary antibody is added. Finally, the plate is treated with an HRP-labeled secondary antibody followed by addition of the HRP substrate to produce chemiluminescence that can then be measured using a chemiluminescence reader.

The NRG1β: HER3 (ERBB3) Chemiluminescent Assay Kit is an ELISA-based assay designed to measure the binding between NRG1β (neuregulin-1β) and HER3 (human epidermal growth factor receptor 3, also known as ERBB3) for screening and profiling applications. The NRG1β: HER3 (ERBB3) Chemiluminescent Assay Kit comes with enough purified NRG1β (amino acids 2-246) and HER3 (amino acids 20-643) proteins, detection antibody, assay buffer, and detection reagent for 100 enzyme reactions. A 96-well plate is coated with NRG1β protein. After coating and blocking, HER3 is added in an optimized assay buffer. Unbound HER3 is washed away, and the plate is incubated with a detection antibody. Finally, ELISA ECL substrate is added to produce chemiluminescence that can be measured using a chemiluminescence reader. The chemiluminescence signal is proportional to the efficacy of HER3 binding to NRG1β.

The NRG1β: HER4 (ERBB4) Chemiluminescent Assay Kit is an ELISA-based assay designed to measure the binding between NRG1β (neuregulin-1β) and HER4 (human epidermal growth factor receptor 4, also known as ERBB4) for screening and profiling applications. The NRG1β: HER4 (ERBB4) Chemiluminescent Assay Kit comes with enough purified NRG1β (amino acids 2-246) and HER4 (amino acids 26-651) proteins, detection antibody, assay buffer, and detection reagent for 100 enzyme reactions. A 96-well plate is coated with NRG1β protein. After coating and blocking, HER4 is added in an optimized assay buffer. Unbound HER4 is washed away, and the plate is incubated with a detection antibody. Finally, ELISA ECL substrate is added to produce chemiluminescence that can be measured using a chemiluminescence reader. The VEGFR1:VEGF165 [Biotinylated] Inhibitor Screening Chemiluminescence Assay Kit is an ELISA designed for screening and profiling molecules that block the binding between VEGFR1 (vascular endothelial growth factor receptor 1, also known as fms related receptor tyrosine kinase 1) and VEGF165 (vascular endothelial growth factor 165). This kit comes in a convenient 96-well format, with enough recombinant biotin-labeled VEGF165 (amino acids 27-191(end)), purified VEGFR1 (amino acids 27-756), streptavidin-labeled HRP, and assay buffer for 100 binding reactions. A 96-well plate is coated with VEGFR1 protein. After blocking, the plate is pre-incubated with an inhibitor or neutralizing antibody. After incubation with Biotin-VEGF165, the plate is washed and Streptavidin-HRP is added. The ELISA ECL substrate is added, and the resulting signal can be measured using a chemiluminescence microplate reader. The chemiluminescence signal is proportional to the binding of VEGFR1 to VEGF165.

The iQuant™ microRNA Assay Kit is ideal for quantification of small RNA, including microRNA and siRNA, and both single stranded and double stranded RNA. The assay kit is highly selective for small RNA over rRNA or large mRNA. The assay kit offers advantages in stability, linear dynamic range, and sensitivity over other traditional of RNA quantitation. The assay kit contains concentrated quantitation reagent, dilution buffer, and pre-diluted microRNA standards. Simply dilute the reagent using the buffer provided, add your sample (any volume between 1 µl and 20 µl is acceptable), and read the fluorescence using fluorescence plate reader or Fluorometer (Figure 1). The assay is well tolerated to common contaminants such as proteins, salts, solvents, detergents, or free nucleotides. The assay can be adapted for use in microplates, tubes or cuvettes.

The Fc (IgG1): FcRn Inhibitor Screening Colorimetric Assay Kit is designed for the screening and profiling of neutralizing antibodies or inhibitors of the interaction between human Fc (IgG1) and human FcRn (Neonatal Fc receptor for IgG). This kit comes in a convenient 96-well or 384-well format, with purified Biotinylated-FcRn complex (Fc receptor amino acids 24-297 and B2M amino acids 21-119) and Fc (IgG1) (amino acids 100-330) proteins, Streptavidin-HRP, and assay buffers for 100 or 400 reactions. A 96-well plate is coated with Fc (IgG1) protein. After blocking, the plate is pre-incubated with an inhibitor or neutralizing antibody. Upon subsequent incubation with FcRn-biotin, the plate is treated with Streptavidin-HRP followed by addition of a colorimetric HRP substrate to produce color, which can be quenched and measured using a UV/Vis microplate reader. The signal is proportional to the binding of FcRn to Fc (IgG1).
