The E. coli in vivo biotinylation kit comes in a convenient, easy-to-use format and contains all the components necessary to label biotin-acceptor peptides with biotin in E. coli. Strain BL21, a chemically competent E. coli B strain. The cells include an IPTG-inducible plasmid containing the BirA gene and the resistance gene for streptomycin or spectinomycin. Just transform the competent cells with a vector expressing your protein of interest with an AviTag™ sequence (example: pAN or pAC vectors). After transformation, BirA expression is induced and biotin is added to the culture. The BirA efficiently catalyzes the specific attachment of biotin to the AviTag.

Protein phosphorylation is one of the most important post-translational modification processes. Phosphorylation is reversibly regulated by Protein Kinases (PKs) and Protein Phosphatases (PTPs). PTP1B (PTPN1) is known to catalyze dephosphorylation of insulin receptor kinases and plays a critical role in insulin signaling. The PTP1B Colorimetric Assay Kit is designed to measure PTP1B activity for screening and profiling applications, in a homogeneous assay with no time-consuming washing steps. The PTP1B Colorimetric Assay Kit is optimized for both continuous and end-point assay formats.

The BRD4 (BD1) Inhibitor Screening Assay Kit is designed to measure the inhibition of BRD4 bromodomain 1 (BD1) from binding to its substrate. The key to the BRD4 (BD1) Inhibitor Screening Assay Kit is the highly specific binding of the BRD4 bromodomain 1 to the acetylated histone substrate. With this kit, only three simple steps on a microtiter plate are required. First, a sample containing BRD4 bromodomain 1 and an inhibitor of choice is incubated with the biotinylated substrate for thirty minutes. Next, acceptor beads are added, then donor beads, followed by reading the Alpha-counts.

The BRD2 (BD2) Inhibitor Screening Assay Kit is designed to measure the inhibition of BRD2 bromodomain 2 (BD2) from binding to its substrate. The key to the BRD2 (BD2) Inhibitor Screening Assay Kit is the highly specific binding of the BRD2 bromodomain 2 to the acetylated histone substrate. With this kit, only three simple steps on a microtiter plate are required. First, a sample containing BRD2 bromodomain 2 and an inhibitor of choice is incubated with the biotinylated substrate for thirty minutes. Next, acceptor beads are added, then donor beads, followed by reading the Alpha-counts.
