Total RNA was prepared from cell line WM3211. WM3211 is a tumorigenic (VGP) primary melanoma cell line with competence for metastasis. These cells display fibroblastic morphology in culture. This cell line was established from a metastatic site (lymph node) in a 34-year-old female. This cell line contains a L576P mutation at position 576 in the c-KIT gene. The L576P mutation results in an amino acid substitution at position 576 in KIT, from a Leucine (L) to a proline (P). This mutation occurs within the juxtamembrane domain. Mutant KIT proteins have increased kinase activity and transforming activity in vitro. This cell line is wild type for BRAF, PTEN, N-RAS, and CDK4. WM3211 cells produce xenograft tumors when injected into immunocompromised mice.

Total RNA was prepared from cell line WM3248. WM3248 is a human melanoma cell line. This cell line features the specific V600E (Val600Glu) mutation at codon 600 in the BRAF gene. This mutation causes constitutively active kinase activity and activation of MEK and ERK signaling pathway. This cell line also expresses PTEN loss of function including mutated and hemizygous PTEN deletion and is wild type for N-RAS, c-KIT, and CDK4.

Total RNA was prepared from cell line WM3268. WM3268 is a tumorigenic (VGP) primary melanoma cell line with competence for metastasis. These cells display mesenchymal morphology in culture. This cell line contains a Q61K mutation at position 61 in the N-RAS gene which causes increased signaling via the extracellular signal-regulated MAPK/ERK kinase pathways to enhance proliferation. WM3268 cells produce xenograft tumors when injected into immunocompromised mice.
