MIC1 LNPs can be used as vehicle to deliver mRNA vaccine to tumor-draining lymph nodes. This LNP exhibited high transfection efficiency and lysosomal escape ability. YHS-12 is an ionizable lipid used to generate lipid nanoparticles to deliver RNA. LNPs optimized with the formula 15:10:25:2.5 (YHS-12/DOPE/cholesterol/DMG-PEG) delivered mRNA and siRNA together to Raw264.7-Luc cells. YHS-12 LNPs incorporating CRV peptide were able to target monocytes in situ to deliver MRSA-targeted chimeric antigen receptor (CAR) mRNA (SasA-CAR mRNA) and CASP11 siRNA. The engineered monocytes showed increased bactericidal activity in a MRSA-induced mouse model.
