Azide-PEG-acid, MW 5,000 is a polyPEG reagent with an azide group with a terminal carboxylic acid. The azide group can react with alkyne, BCN, DBCO via Click Chemistry. The terminal carboxylic acid can react with primary amine groups in the presence of activators (e.g. HATU). The 5k PEG polymer increases the water-solubility of the molecule.

Azido-PEG-NHS ester, MW 2,000 (N3-PEG-SPA, MW 2,000) is a Click chemistry-reactive PEG active ester containing an azide group. Azido-PEG-NHS ester (N3-PEG-SPA) offers several benefits over the Azido-PEG-CH2CO2-NHS (N3-PEG-SCM). Azido-PEG-NHS ester is more stable in aqueous solutions compared to Azido-PEG-CH2CO2-NHS. It is a better choice for conjugations that are to be performed in aqueous solutions. Azido-PEG-NHS ester (N3-PEG-SPA) is also more stable for storage than Azido-PEG-CH2CO2-NHS (N3-PEG-SCM). Azido-PEG-NHS ester is reactive with primary amine but at a lower rate compared to Azido-PEG-CH2CO2-NHS.
