Interleukin 17 receptor A, also known as IL17RA and CDw217 (cluster of differentiation w217), is a human gene. The IL17R gene is mapped to 22q11.22-q11.23 by radiation hybrid analysis. Interleukin 17A (IL17A) is a proinflammatory cytokine secreted by activated T-lymphocytes. It is a potent inducer of the maturation of CD34-positive hematopoietic precursors into neutrophils. The protein encoded by this gene is a ubiquitous type I membrane glycoprotein that binds with low affinity to interleukin 17A. Interleukin 17A and its receptor play a pathogenic role in many inflammatory and autoimmune diseases such as rheumatoid arthritis. Like other cytokine receptors, this receptor likely has a multimeric structure. Also, it is showed that mouse Il17r bound HSV-13 and mouse Il17. Recombinant soluble Il17r inhibited mitogen-induced T-cell proliferation and Il2 production.

IL17RB, also known as CRL4, is a protein that in humans is encoded by the IL17RB gene. It is located on 3p21.1. IL17RB is produced by activated T cells exhibiting proinflammatory activities. The protein encoded by this gene is a cytokine receptor. This receptor specifically binds to IL17B and IL17E (IL25), but does not bind to IL17 (A) or IL17C. Interleukin 17D (IL-17D) is a member of the unique IL-17 family, which share a conserved cysteine motif. It is mapped to 13q12.11. IL-17D regulates cytokine production in endothelial cells and inhibits hemopoiesis in vitro. Purified recombinant IL-17D inhibited myeloid progenitor cell colony formation. IL-17D can enhance endothelial cell production of IL6, IL8, and GMCSF, but not other cytokines. And it is preferentially expressed in skeletal muscle, brain, adipose tissue, heart, lung, and pancreas.

Interleukin-25 (IL-25), also known as interleukin-17E (IL-17E), is a protein that in humans is encoded by the IL25 gene. It is a cytokine that belongs to the IL-17 cytokine family and mapped to 14q11.2. IL-25 is secreted by type 2 helper T cells (Th2) and mast cells. It has been found that IL-25 can induce Th2-type cytokine expression and production by non-T, non-B accessory cells that express high levels of major histocompatibility complex class II and low levels of Cd11c. This cytokine also can induce NF-kB activation, and stimulate the production of IL8. What’s more, IL-25 is an important molecule controlling immunity of the gut and has been implicated in chronic inflammation associated with the gastrointestinal tract.
