N-Boc-N-bis(PEG4-Propargyl) is a bifunctional PEG compound containing two terminal alkynes that are combined together at a Boc-protected secondary amine. Terminal alkynes are most commonly used in copper click chemistry with azides to form stable triazoles with the target molecule. The secondary amine may be deprotected under acidic conditions to allow for alkylation at that position, increasing the molecule’s structural complexity. The use of a central amine also allows for hydrogen bonding, further increasing this compound’s water solubility.

N-(Mal-PEG4)-N-bis(PEG4-N-Boc) is a trifunctional linker with two Boc-protected amines and a maleimide group. Maleimide is a site-specific covalent linker which easily reacts with thiol groups via a simple Michael addition. The amines can be easily deprotected using acidic conditions to allow for coupling with carboxylic acids in the presence of an activator like HATU. The junction combining these functional groups is a tertiary amide, which is highly water soluble through hydrogen bonding effects.

Lipid A6 is an ionizable amino lipid featuring a dimethyl headgroup linked by an ester to a central branched linker featuring two unsaturated esters with internal alkynes. Its structure is similar to D-Lin-MC3-DMA, however it has shown a significantly improved ability in cell transfection over the respective MC3/C12-200/cKK-E12 LNPs.

304-O13 exhibits comparable potency to the non-degradable C12-200 standard while displaying significantly reduced toxicity at high doses. It's important to mention that ester-containing ionizable lipids usually show lower effectiveness compared to non-degradable counterparts due to rapid hydrolysis hindering efficient delivery.

Amine-PEG8-Amido-tri-(carboxyethoxymethyl)-methane, HCl salt is a tetrafunctional PEG linker including three terminal caboxyl groups, which can react with primary amine groups in the presence of activators such as HATU, and terminal amine for coupling with an NHS ester to form an amide bond. The PEG8 linker improves the hydrophilicity of the compound.
