The Homogeneous full length SHP-2 Assay Kit is a complete assay system designed to measure full length SHP-2 activity for screening and profiling applications. Using this kit, only two simple steps on a microtiter plate are needed to analyze the full length SHP-2 activity level. At the first step, the SHP-2 enzyme is preincubated with SHP-2 Activating Peptide to activate the enzyme. At the second step, the fluorogenic substrate, DiFMUP, is added in the mixture and the enzymatic activity releases DiFMU fluorophore that can then be measured using a fluorescence reader.

The FTO Chemiluminescent Assay Kit is designed to measure FTO activity for screening and profiling applications. The key to the FTO Chemiluminescent Assay Kit is a highly specific antibody that recognizes methylated substrate. Signal is inversely related to the activity. With this kit, only three simple steps on a microtiter plate are required for detection of demethylase activity. First, FTO enzyme is incubated with the substrate. Next, primary antibody is added. Finally, the plate is treated with an HRPlabeled secondary antibody followed by the addition of the HRP substrate to produce chemiluminescence that can be measured using a chemiluminescence reader.

The NSD2 Chemiluminescent Assay Kit is designed to measure NSD2 activity for screening and profiling applications. The key to the NSD2 Chemiluminescent Activity Assay Kit is a highly specific antibody that recognizes methylated residue of Histone H3. With this kit, only three simple steps are required for methyltransferase detection. First, Sadenosylmethionine is incubated with a sample containing assay buffer and methyltransferase enzyme. Next, primary antibody is added. Finally, the plate is treated with an HRP-labeled secondary antibody followed by addition of the HRP substrate to produce chemiluminescence that can then be measured using a chemiluminescence reader.

The CSF1R SRE Reporter Kit is designed for monitoring the activity of the CSF1R signaling pathway in cultured cells. The kit contains a transfection-ready vector for CSF1R and SRE luciferase reporter vector. Upon ligand binding, active CSF1R will initiate the MAPK/ERK signaling pathway, leading to expression of the SRE-controlled reporter. This reporter contains the firefly luciferase gene under the control of multimerized SRE responsive elements located upstream of a minimal promoter. The SRE reporter is premixed with a constitutively-expressing Renilla luciferase vector that serves as an internal control for transfection efficiency. The kit also includes a non-inducible firefly luciferase vector premixed with constitutively- expressing Renilla luciferase vector as a negative control. The non-inducible luciferase vector contains the firefly luciferase gene under the control of a minimal promoter, without any additional response elements. The negative control is critical for determini

Sortases catalize a transpeptidation reaction, where the enzyme cleaves the amide bond between the threonine and glycine of the sortase recognition sequence (LPXTG for S. aureus Sortase A), generating a thioacyl intermediate. Subsequently, this intermediate is resolved by the N-terminus of an oligoglycine nucleophile, creating a new peptide bond that links the substrate to the incoming nucleophile. The Sortagging Kit takes advantage of this reaction to label virtually any recombinant antibody or protein containing the sortase recognition sequence. Labeling efficiency of greater than 90% is achieved by taking advantage of the highly active Sortase 5Δ Pentamutant. Simply combine your protein of interest with Sortase 5Δ and a polyglycine label, and incubate for 1 hour at 37°C. Purification columns are included to remove Sortase 5Δ and excess label. The kit also includes Ubiquitin-LPETGH6 and GGG-Clover as positive controls.

The recombinant expression vectors are designed to express human engineered T cell receptor (TCR) activator and woodchuck (groundhog, Marmota monax) PD-L1 (GenBank Accession #HQ403651) in mammalian cells. The transfected cells can be used in conjunction with Woodchuck PD-1/NFAT Reporter/Jurkat T cells (BPS #79456) to study the interactions of PD-1 with PD-L1 ligand in a cellular context and screen for modulators of this signaling pathway.

The Tyrosine Phosphorylation Assay Kit is designed to measure TYR phosphorylation of proteins. The key to the Tyrosine Phosphorylation Assay Kit is a specific antibody mixture that provides sequence-independent recognition of phosphor-TYR residues. With this kit, only three simple steps on a microtiter plate are required for detection of the phosphorylation state. First, the protein of interest is coated on the strip plate. Next, primary antibody is added. Finally, the strips are treated with an HRP-labeled secondary antibody followed by addition of the HRP substrate to produce chemiluminescence that can then be measured using a chemiluminescence reader.

Notch Pathway Reporter kit is designed for monitoring the activity of the Notch signaling pathway in cultured cells. The kit contains a transfection-ready expression vector for Human NOTCH1 that has a deletion of the entire extracellular domain, leaving the transmembrane and intracellular domains intact (Notch1DE). Inside the cells, the NOTCH1 DE is cleaved by γ-secretase and active NOTCH1 NICD is released into the nucleus. The kit also contains CSL (CBF1/RBP-Jk) luciferase reporter vector, which is a Notch pathway-responsive reporter. This reporter contains the firefly luciferase gene under the control of multimerized CSL responsive elements upstream of a minimal promoter. The CSL (CBF1/RBP-Jk) reporter is premixed with constitutively expressing Renilla (sea pansy) luciferase vector, which serves as an internal positive control for transfection efficiency. The kit also includes a non-inducibl
