The CD27:CD70[Biotinylated] Inhibitor Screening Assay Kit is designed for screening and profiling inhibitors of CD27:CD70 signaling. This kit comes in a convenient 96- well format, with biotin-labeled CD70, purified CD27, streptavidin-labeled HRP, and assay buffer for 100 binding reactions. The key to this kit is the high sensitivity of detection of biotin-labeled CD70 by streptavidin-HRP. Only a few simple steps on a microtiter plate are required for the assay. First, CD27 is coated on a 96-well plate. Next, CD70 is incubated with CD27 on the plate. Finally, the plate is treated with streptavidin-HRP followed by addition of an HRP substrate to produce chemiluminescence, which can be measured using a chemiluminescence reader.

Histone acetyltransferase p300 (also known as KAT3B) is involved in various cellular events, and its dysfunction is linked to a number of human diseases including Parkinson's, Huntington's, and Alzheimer's diseases. The P300 Chemiluminescent Assay Kit is based on the p300 enzyme transferring an acetyl group from an acetyl donor (acetyl CoA) to a histone substrate. The acetylated histone is recognized by a highly specific primary antibody, followed by an HRP-labeled secondary antibody. The chemiluminescence produced by HRP can be measured using a chemiluminescence reader.

Sortases catalyze a transpeptidation reaction, where the enzyme cleaves the amide bond between the threonine and glycine of the sortase recognition sequence (LPXTG for S. aureus Sortase A), generating a thioacyl intermediate. Subsequently, this intermediate is resolved by the N-terminus of an oligoglycine nucleophile, creating a new peptide bond that links the substrate to the incoming nucleophile. Sortase variants are available that may exhibit enhanced activity with the substrate of interest. This kit provides wildtype Sortase and four different mutants to evaluate which sortase is best for the chosen application.
