UBCH13 is a noncanonical ubiquitin conjugating enzyme (E2) that has been implicated in a variety of cellular signaling processes due to its ability to catalyze the formation of lysine 63-linked polyubiquitin chains on various substrates. In particular, UBCH13 is required for signaling by a variety of receptors important in immune regulation, making it a candidate target for inflammatory diseases. UBCH13 is also critical for double-strand DNA repair and thus a potential radiosensitizer and chemosensitizer target for oncology. The UBCH13 TR-FRET Assay Kit is designed to measure UBCH13 ubiquitination activity in a homogeneous 384 reaction format. It utilizes biotin-labeled ubiquitin and a terbium-labeled anti-His antibody to complete the TR-FRET pairing. This FRET-based assay requires no time-consuming washing steps, making it especially suitable for high throughput screening applications.

The TNFR2:TNF-alpha[Biotinylated] Inhibitor Screening Assay Kit is designed for screening and profiling inhibitors of TNFR2:TNF-alpha. The key to this kit is the high sensitivity of detection of biotin-labeled TNF-alpha by streptavidin-HRP. Only a few simple steps on a microtiter plate are required for the assay. First, TNFR2 is coated on a 96-well plate. Next, biotinylated TNF-alpha is incubated with TNFR2 on the plate. Finally, the plate is treated with streptavidin-HRP followed by addition of an HRP substrate to produce chemiluminescence, which can be measured using a chemiluminescence reader.

The MLL4 Complex Chemiluminescent Assay Kit is designed to measure MLL4 activity for screening and profiling applications, using purified MLL4 and its complex components: WDR5, RbBP5, Ash2, and DPY30. The key to the MLL4 Complex Chemiluminescent Assay Kit is a highly specific antibody that recognizes methylated K4 residue of Histone H3. With this kit, only three simple steps on a microtiter plate are required for methyltransferase detection. First, S-adenosylmethionine is incubated with a sample containing assay buffer and methyltransferase enzyme. Next, primary antibody is added. Finally, the plates are treated with an HRP-labeled secondary antibody followed by the addition of the HRP substrate to produce chemiluminescence that can be measured using a chemiluminescence reader.

Protein phosphorylation is one of the most important post-translational modification processes. Phosphorylation is reversibly regulated by Protein Kinases (PKs) and Protein Phosphatases (PTPs). PTP1B (PTPN1) is known to catalyze dephosphorylation of insulin receptor kinases and plays a critical role in insulin signaling. The Fluorogenic PTP1B (Catalytic Region) Assay Kit is designed to inhibitors of the catalytic region of PTP1B in a homogeneous assay with no time-consuming washing steps. Note: To identify inhibitors of the regulatory region of PTP1B, please use our Fluorogenic PTP1B (Full Length) Assay Kit, #79766).

The PROTAC Optimization Kit for BET Bromodomain-Cereblon Binding is designed for testing and profiling of PROTACs directed against BET Bromodomain family and Cereblon complex. Cereblon (CRBN) is a Substrate recognition component of a DCX (DDB1-CUL44-Rbx1) E3 protein ligase complex that mediates the ubiquitination and subsequent proteasomal degradation of target proteins. The PROTAC Optimization Kit for BET Bromodomain-Cereblon Binding comes in a convenient AlphaLISA® format, with dBET1 PROTAC, Immuno Buffer 1, and purified BRD3(BD2) and CRBN for 384 reactions. (+)JQ1 is included as a control inhibitor of PROTAC binding to BRD3(BD2). With this kit, only three simple steps on a microtiter plate are required for PROTAC activity detection. First, a sample containing PROTAC is incubated with CRBN and BRD3(BD2), one of BET bromodomains. Next, acceptor beads are added, then donor beads, followed by reading the Alpha-counts.
