The HPK1 Assay Kit is designed to measure HPK1 activity for screening and profiling applications using Kinase-Glo® MAX as a detection reagent.

The recombinant expression vectors are designed to express human engineered T cell receptor (TCR) activator and mouse PD-L1 (GenBank Accession #NM_021893) in mammalian cells. The transfected cells can be used in conjunction with mouse PD-1/NFAT Reporter/Jurkat T cells (BPS Bioscience #79762) to study the interactions of PD-1 with PD-L1 ligand in a cellular context and to screen for modulators of this signaling pathway. Functional Validation and Assay Performance: In this assay, mouse PD-1/NFAT Reporter/Jurkat T cells are used as effector cells; HEK293 cells transiently transfected with the PD-L1 vector over-express mouse PD-L1 and an engineered T cell receptor (TCR) activator and are used as target cells. When these two cells are co-cultivated, TCR complexes on effector cells are activated by TCR activator on target cells, resulting in expression of the NFAT luciferase reporter. The VSIG-3:VISTA[Biotinylated] Inhibitor Screening Assay Kit is designed for screening and profiling inhibitors of VISTA binding to VSIG-3. This kit comes in a convenient 96-well format, with biotin-labeled VISTA, purified VSIG-3, streptavidin-labeled HRP, and assay buffer for 100 binding reactions. The key to this kit is the high sensitivity of detection of biotin-labeled VISTA by streptavidin-HRP. Only a few simple steps on a microtiter plate are required for the assay. First, VSIG-3 is coated on a 96-well plate. Next, biotinylated VISTA is incubated with VSIG-3 on the plate. Finally, the plate is treated with streptavidin-HRP followed by addition of an HRP substrate to produce chemiluminescence, which can be measured using a chemiluminescence reader.

Recent evidence suggests a critical role for c-Cbl in angiogenesis and human solid organ tumors, making it a potential drug target in cancer immunotherapy. The c-CBL TR-FRET Assay Kit is designed to measure c-CBL auto-ubiquitination activity in a homogeneous 384 reaction format. It utilizes Biotin-labeled Ubiquitin and a terbium-labeled anti-c-CBL antibody to complete the TR-FRET pairing. This FRET-based assay requires no time-consuming washing steps, making it especially suitable for high throughput screening applications.

The LAG3:FGL1[Biotinylated] Inhibitor Screening Assay Kit is designed for screening and profiling inhibitors of LAG3:FGL1. The key to this kit is the high sensitivity of detection of biotin-labeled FGL1 by streptavidin-HRP. Only a few simple steps on a microtiter plate are required for the assay. First, LAG3 is attached on a nickel-coated 96-well plate. Next, biotinylated FGL1 is incubated with LAG3 on the plate. The PROTAC Optimization Kit for BET Bromodomain-Von Hippel Lindau (VHL) Binding is designed for testing and profiling PROTACs directed against the BET Bromodomain family and VHL complex. VHL is a substrate recognition component of E3 protein ligase complex and is linked by Elongin C to a heterodimeric Cul2/Rbx1 module, which functions as a potent activator of the ubiquitination of target proteins by an E2 conjugating enzyme. Elongin B interacts with the complex through Elongin C and stabilizes the binding of Elongin C to VHL. Mutations in VHL are associated with the inherited von Hippel-Lindau cancer syndrome and numerous forms of renal cell carcinoma. With this kit, only three simple steps on a microtiter plate are required for PROTAC activity<br />detection. First, a sample containing PROTAC is incubated with VHL and BRD3(BD2), one of the<br />BET bromodomains. Next, acceptor beads are added, then donor beads, followed by reading the Alpha-counts.
