5(6)-Carboxyrhodamine 110 NHS Ester is the nonsulfonated analog of the BP Fluor 488 dye. The amine-reactive 5(6)-Carboxyrhodamine 110 NHS Ester can be used to create bright and photostable green-fluorescent bioconjugates with excitation/emission maxima ~502/527 nm. The conjugates of this dye often used for Fluorescence Correlation Spectroscopy (FCS). For many applications, 5(6)-Carboxyrhodamine 110 NHS Ester is preferred over 5-(6)-carboxyflu Reagent grade, for research purpose. Please contact us for GMP-grade inquiries.

Although the mixed isomers of 5(6)-TAMRA Acid is a preferred, routinely used orange-fluorescent dye for staining proteins, it is rearly used for labeling peptides and nucleotides. Purification of 5(6)-TAMRA labeled peptide and nucleotides might be troublesome due to significant signal broadening in HPLC purification. Peptides and nucleotides labeled with a single isomer TAMRA usually give better resolution in HPLC purification that is often required in the conjugation processes. 5-TAMRA is more often used for labeling peptides and proteins. 6-TAMRA is predominately used for labeling nucleotides and sequencing nucleic acids. 5-TAMRA dye is a bright fluorescent label is compatible with various excitation sources including mercury arc, tungsten and xenon arc lamps, the 544 nm line of the Helium-Neon laser and the 532 nm green laser line. 5(6)-TAMRA NHS Ester (5(6)-Carboxytetramethylrhodamine, TMR, TRITC) is a bright orange-fluorescent dye with excitation ideally suited to the 532 nm or 546 nm laser lines. It has been used widely for preparing peptide, protein, nucleotide and nucleic acid conjugates, especially fluorescent antibodies and avidin derivatives used in immunochemistry. The absorbance and emission maxima of TAMRA conjugates are 553 nm and 575 nm respectively. TAMRA NHS Ester reacts specifically and efficiently with a primary amine (e.g., side chain of lysine residues or aminosilane-coated surfaces) at pH 7-9 to form a stable, covalent amide bond. The NHS ester (or succinimidyl ester) is the most popular tool for conjugating dyes to the primary amines of protein or antibody (Lys), amine-modified oligonucleotides, and other amine-containing molecules. 5-TAMRA cadaverine can used to modify carboxylic acid group in the presence of activators (e.g. EDC, or DCC) or activated esters (e.g. NHS esters) through a stable amide bond. It also can be reversibly coupled to aldehydes and ketones to form a Schiff base – which can be reduced to a generate stable amine derivative by sodium borohydride (NaBH4) or sodium cyanoborohydride (NaCNH3). Although the mixed isomers of 5(6)-TAMRA cadaverine is a preferred, routinely used orange-fluorescent dye for staining proteins, it is rearly used for labeling peptides and nucleotides. Purification of 5(6)-TAMRA labeled peptide and nucleotides might be troublesome due to significant signal broadening in HPLC purification. TAMRA Azide, isomer 5 (also known as Tetramethylrhodamine 5-Carboxamido-(6-Azidohexanyl)) is the orange-fluorescent probe that is compatible with various excitation sources including mercury arc, tungsten and xenon arc lamps, the 544 nm line of the Helium-Neon laser and the 532 nm green laser line. It is predominantly used for detection of terminal alkyne-tagged biomolecules via a copper-catalyzed click reaction (CuAAC). It also reacts with strained cyclooctyne via a copper-free “click chemistry” reaction to form a stable triazole and does not require Cu-catalyst or elevated temperatures. For the detection of low abundance targets or where significant increase in signal intensity is desired please consider using next generation azides probes containing an internal copper-chelating motif. TAMRA Picolyl Azide is an advanced fluorescent probe that incorporatess a copper-chelating motif to raise the effective concentration of Cu(I) at the reaction site. The rate of CuAAC reaction depends on concentrations of all reagents, including copper, thus the raise of the effective copper concentration at the reaction site dramatically increase the rate of CuAAC reaction without the need to increase concentration of azide reagent and copper. This probe is compatible with various excitation sources including mercury arc, tungsten and xenon arc lamps, the 544 nm line of the Helium-Neon laser and the 532 nm green laser line. In addition, the use of TAMRA Picolyl Azide instead of conventional TAMRA Azide allows for at least a tenfold reduction in the concentration of the copper catalyst without sacrificing the efficiency of labeling. In summary, the introduction of a picolyl moiety into an azide probe leads to a substantial increase in the sensitivity of alkyne detection. This will be of
