The PDE6C Assay Kit is a fluorescence polarization (FP), homogeneous, 96-well assay kit designed for the screening and profiling of PDE6C (Phosphodiesterase 6C) inhibitors. This assay takes advantage of a specific fluorescent phosphate-binding nanoparticle. The kit contains enough purified recombinant PDE6C, fluorescent probe, PDE assay buffer, Binding Agent, and diluent for 100 reactions. The assay uses a fluorescein-labeled cyclic guanosine monophosphate (cGMP-FAM for PDE6C), in which the phosphate group is engaged within the cyclic nucleotide. This is a very small molecule that rotates fast (low FP). PDE6C catalyzes the hydrolysis of the phosphodiester bond in the cyclic nucleotide and frees the phosphate group. In a second step the free phosphate group is recognized by a specific phosphate-binding nanobead (Binding Agent) leading to the formation of a large complex, with restricted movement (high FP). The FcRn:HSA Binding Chemiluminescent Assay Kit is designed for screening and profiling neutralizing antibodies or blockers of the interaction between Human Serum Albumin (HSA) and human FcRn. This kit comes in a convenient 384-well format, with Biotinylated HSA and FcRn (FCGRT/B2M) (amino acids 24-297 of FCGRT and 21-119 of B2M), Streptavidin-HRP, and assay buffers for 400 reactions. The assay requires only a few steps. First, FcRn is coated on a 384-well plate overnight. After blocking, the protein is pre-incubated with the neutralizing antibody or blocker. Upon subsequent incubation with Biotin-HSA, the plate is treated with Streptavidin-HRP followed by addition of an HRP substrate to produce chemiluminescence, which can then be measured using a chemiluminescence reader.

The TSLPR:TSLP [Biotinylated] Small Molecule Inhibitor Screening Chemiluminescence Assay Kit is designed for screening and profiling small molecules that block the binding between TSLPR (thymic stromal lymphopoietin receptor) and TSLP. This kit comes in a convenient 96-well format, with enough recombinant human biotin-labeled TSLP (amino acids 29-159), human TSLPR (amino acids 25-231), streptavidin-HRP, and assay buffer for 100 reactions. A 96-well plate is coated with TSLPR protein. After coating and blocking, TSLP is added in an optimized assay buffer. Unbound TSLP is washed away, and the plate is incubated with streptavidin-HRP. Finally, ELISA ECL substrate is added to produce chemiluminescence that can be measured using a chemiluminescence reader.
