The PLPro (Papain-Like Protease) substrate is a small 5-mer fluorogenic peptide (Z- RLRGG-AMC) used to measure the activity of SARS family PLPro proteases. Release of the C-terminal AMC dye (7-amido-4-methylcoumarin ) by proteolysis generates fluorescence, which can be monitored at an excitation wavelength of 345 nm and an emission wavelength of 445 nm. Conditions optimized for use with: Papain-like Protease (SARS-CoV-2) Assay Kit: Protease Activity (BPS Bioscience #79995). The peptide corresponds to a short sequence of ubiquitin and may also be used as substrate of USP5 (isopeptidase T) and ubiquitin C-terminal hydrolases (UCH family, for example UCHL1).

The pandemic coronavirus disease 2019 (COVID-19) is caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). The Spike glycoprotein is expressed on the surface of the virus as a trimer. Each Spike protein consists of two subunits, S1 and S2, and the S1 subunit has a receptor binding domain (RBD) which recognizes and attaches to the Angiotensin-Converting Enzyme 2 (ACE2) receptor found on the surface of type I and II pneumocytes, endothelial cells, and ciliated bronchial epithelial cells. Drugs targeting the interaction between the Spike protein of SARS-CoV-2 and ACE2 may offer some protection against the viral infection. The SARS-CoV-2 Spike Trimer (S1+S2) :ACE2 Inhibitor Screening Assay Kit includes the Spike protein in its native trimeric conformation to provide a more physiologically relevant screen for inhibitors of the Spike S1:ACE2 interaction. The SARS-CoV-2 Spike Trimer (S1+S2) :ACE2 Inhibitor Screening Assay Kit is designed for screening and profiling inhibitor

The Fluorogenic Prolyl OligoPeptidase (POP) Assay Kit is a complete assay system designed to measure activity of the purified POP enzyme. The Fluorogenic POP Activity Kit eliminates the dealing with radioactive materials and chromatography in traditional assays. Purified human recombinant POP is included in the kit as a positive control. Using this kit, only one simple step, in which the fluorometric substrate is incubated with purified POP, is needed to analyze the POP activity level. The resulting fluorescent product can then be easily measured with a microtiter-plate fluorimeter.
