Linolenoyl LPA-d5 is a deuterated analog of 18:3 LPA labeled on the glycerol backbone. Lysophosphatidic Acid (LPA) is a small lysophospholipid involved in diverse cellular processes such as cell proliferation chemotaxis platelet aggregation wound healing angiogenesis tumor invasion and smooth muscle contraction. LPA binds to several G-coupled protein receptors to initiate its biological functions. In cancer LPA primarily promotes cell survival migration and invasion.

Phosphatidylcholine (PC) is generally the most abundant lipid in animal cell membranes providing structural framework. PC is more common in the outer leaflet where it functions as part of the permeability barrier. PC is also the primary substrate of Phospholipase D enzymes which produce the signaling lipids-phosphatidic acid and lysophosphatidic acid. This analog is deuterated on the choline headgroup.

LysoPC is generated through hydrolysis of the sn-2 acyl chain of PC by phospholipase A2. It is a major component of oxidized low-density lipoprotein and is believed to have an important role in inflammatory diseases and atherosclerosis. LysoPC is the substrate for Autotaxin/LysoPLD generating lysophosphatidic acid which has an important role in cell motility. This analog is deuterated on the choline headgroup.

Ionizable lipids in combination with other lipids make up the lipid nanoparticles (LNPs) which are used to deliver RNA-based therapeutics. LNPs incorporating Lipid 5 (from Sabnis et al.) demonstrated potent delivery of mRNA in primates but also were rapidly cleared from the body resulting in low toxicity. Lipid 5 was better at I.V. delivery to the liver while the closely related SM-102 (cat# N-1102) was optimized for intramuscular administration. This is a reagent grade product for research use only.References:1) Sabnis S Kumarasinghe ES Salerno T et al. DLin-MC3-DMA is an ionizable lipid used in combination with other lipids for encapsulation of RNA and DNA into lipid nanoparticles (LNPs). Administration of DLin-MC3-DMA based LNPs have delivered siRNA and reduced Factor VII levels in mice. Delivery of BCR-ABL siRNA reduced leukemic burden in a mouse model of chronic myeloid leukemia (CML).This product is for research use only and not for human use.
