Lactate dehydrogenase (LDH) exists ubiquitously in the cytoplasm of living mammalian cells and the measurement of cytoplasmic LDH activity is a well-accepted assay to quantify living cell numbers and indicate cell viablity. The DHL™ Cell Viability and Proliferation Assay Kit provides researchers with a convenient one-solution and one-step assay to count living cells in a culture and continuously monitor cell proliferation over time by measuring cytoplasmic LDH activity. In this kit, resazurin is used as a sensitive fluorogenic indicator. Resazurin is converted to the strongly fluorescent resorufin (Ex/Em=560nm/590 nm) by cytoplasmic LDH. The kit can detect as few as 48 living cells with a linear range up to 5X104 cells (r2=0.99). This kit can also be used for high throughput screening of cell proliferation or cytotoxicity effect of a variety of compounds.

The measurement of released cytoplasmic lactate dehydrogenase (LDH) is a well-accepted assay to estimate cell membrane integrity and quantify cytotoxicity. The SensoLyte® Cell Cytotoxicity Assay Kit uses resazurin as a sensitive fluorogenic indicator (Ex/Em=560 nm/590 nm upon conversion) to measure LDH activity. The assay can be performed in a mixed population of damaged and viable cells, but it only measures the LDH released from damaged cells. The cytoplasmic LDH in living cells produces little signals under assay condition. There is no need for extra steps to separate living cells and supernatant. The fluorescent signal is proportional to the number of damaged cells (up to 2.5X104 cell, r2>0.95) with the detection limit reaching 100 dead cells. The kit is suitable for high throughput screening of cytotoxicity of a variety of compounds. 384-well or 1536-well format can be used with minor modifications.

Calcein AM (calcein acetoxymethyl ester) is a non-fluorescent reagent that easily permeates cellular membrane. Once it permeates live cells, esterases remove the acetomethoxy group allowing calcein to bind to calcium, which results in a strong green fluorescence. The increase of fluorescence is proportional to the number of live cells. Inside dead cells, the non-fluorescent calcein AM remains intact due to the lack of active esterases. The fluorescent signal from the conversion of calcein AM to calcein can be monitored at Ex/Em=494nm /520 nm. The SensoLyte® Calcein Cell Viability Assay Kit provides a convenient tool to detect live cells. This kit can be used in proliferation, cytotoxicity, invasion, adhesion, migration and other cell-based assays.
