Functional Assays

The CD200:CD200R1 [Biotinylated] Inhibitor Screening Chemiluminescence Assay Kit is designed for screening and profiling molecules that block the binding between CD200 (cluster of differentiation 200) and CD200R1 (CD200 receptor 1). This kit comes in a convenient 96-well format, with enough recombinant human biotin-labeled CD200R1 (amino acids 29-265), human CD200 (amino acids 31-232), streptavidin-HRP, and assay buffer for 100 binding reactions. CD200 is coated onto a 96-well plate. After blocking, the plate is pre-incubated with an inhibitor or neutralizing antibody. After incubation with Biotin-CD200R1, the plate is washed and Streptavidin-HRP is added. After the  ELISA ECL substrate is added  the resulting signal can be measured using a chemiluminescence microplate reader. The chemiluminescence signal is proportional to the binding of CD200 to CD200R1.

The Human NK Cell Isolation Kit is designed to magnetically separate NK cells from a complex immune cell population. This kit is optimized for the isolation of CD56⁺CD3^- cells from normal human peripheral blood mononuclear cells (PBMCs). Cells are incubated with a mix of antibodies followed by conjugation to magnetic beads. They are then placed on a magnet for quick and easy separation. When placed on the magnet, non-NK cells will be immobilized along the side of the tube while NK cells will remain in suspension and can be easily removed for downstream applications.

The PROTAC-Driven Ubiquitination Assay Kit for BET Bromodomains is designed for the testing and profiling of PROTACs targeting the bromodomains on the BET (bromodomain and extraterminal) protein family. After tertiary complex formation between the E3 protein, the PROTAC and the protein of interest, ubiquitination of the target protein is a critical step preceding its degradation. The Cytotoxicity Dye Kit (CFSE, 7-AAD) is a kit designed to determine the cytotoxicity profile of effector cells towards target cells of interest. It consists of CFSE (carboxyfluorescein succinimidyl ester), used to identify target cells in a mixed cell population of target cells and PBMC, and 7-AAD (7-aminoactinomycin D) to label dead cells. CFSE and 7-AAD labeling enables cell cytotoxicity detection at the single-cell level using flow cytometry. The kit also contains Assay Diluent.
