CleanCap® Reagent AG (3' OMe) is TriLink’s proprietary co-transcriptional capping reagent for in vitro transcription of 5’ capped mRNA resulting in a Cap 1 structure. CleanCap has shown to provide up to 98% capping efficiency. CleanCap Reagent AG (3' OMe) includes the 5' N7-Methyl-3'-O-Methylguanosine commonly found in mRNA capped using ARCA, whereas CleanCap Reagent AG has a naturally occurring 5' N7-Methylguanosine structure. PLEASE NOTE: TriLink recommends trying both CleanCap Reagent AG and CleanCap Reagent AG (3’ OMe) whenever possible. CleanCap Reagent AG and CleanCap Reagent AG (3’ OMe) require an AG initiator. To learn more, please review the Product Insert below. Our CleanCap reagents are available in “Research Use” grade as well as GMP-grade “For Further Processing.” GMP product is manufactured in accordance with ISO 9001:2015 and Quality Standards as defined in a GMP Quality Agreement, and must be further processed by the client.

Trusted TriLink quality, exceptional price. A key step in cellular mRNA processing is the addition of a 5’ cap structure, which is a 5'-5' triphosphate linkage between the 5' end of the RNA and a guanosine nucleotide. The cap is methylated enzymatically at the N-7 position of the guanosine to form mature mCAP. When preparing synthetic mRNA, the cap is often added prior to use in order to stabilize the mRNA and significantly enhance translation. Using a 4:1 mixture of a cap analog to GTP in transcription reactions will cap 80% of the resulting mRNAs. If mCAP (pre-methylated CAP) is used, only half of the asymmetrical mCAP will insert in the functional orientation. ARCA can only insert in the correct orientation, resulting in mRNAs that are translated twice as efficiently. TriLink is the least expensive source of high quality ARCA. We also offer CAP and mCAP for more traditional capping reactions.

Pseudouridine (5-ribosyluracil) was the first modified ribonucleoside discovered. It is the most abundant natural modified RNA base, and has been deemed the "fifth nucleoside" in RNA. It can be found in structural RNAs, such as transfer, ribosomal and small nuclear RNA. Pseudouridine has been found to enhance base stacking and translation. Pseudouridine-5'-triphosphate (Pseudo-UTP) is used to impart desirable mRNA characteristics such as increased nuclease stability, increased translation or altered interaction of innate immune receptors with in vitro transcribed RNA. Pseudo-UTP, along with 5-methylcytidine-5'-triphosphate (5-methyl-CTP) has shown innate immune suppression in culture and in vivo while enhancing translation in recent publications. For example, Warren et al. determined an efficient means of reprogramming multiple human cell types using modified mRNA that can express the four primary reprogramming proteins. These cells are referred to as induced pluripotency stem cells (i
