Recombinant Murine sRANK Ligand (CHO derived) (Legacy Tebubio ref. 167315-11C). RANKL and RANK are members of the TNF superfamily of ligands and receptors that play an important role in the regulation of specific immunity and bone turnover. RANK (receptor) was originally identified as a dendritic cell-membrane protein, which, by interacting with RANKL, augments the ability of dendritic cells. These dendritic cells then stimulate naive T-cell proliferation in a mixed lymphocyte reaction, promote the survival of RANK+ T-cells, and regulate T-cell-dependent immune response. RANKL, which is expressed in a variety of cells, including osteoblasts, fibroblasts, activated T-cells and bone marrow stromal cells, is also capable of interacting with a decoy receptor called OPG. Recombinant Murine LIGHT (Legacy Tebubio ref. 167315-12). LIGHT belongs to the TNF family of ligands, and can signal through the herpes virus entry mediator type A receptor (HVEM, TNFRSF14), LTbetaR, or bind to a decoy receptor, DcR3. It is expressed in splenocytes, activated PBL, CD8+ tumor infiltrating lymphocytes, granulocytes, and monocytes. LIGHT has the ability to activate NF-kappaB, to co-stimulate the activation of lymphocytes and to induce apoptosis in certain human tumor cells. APRIL, along with another TNF family member, BAFF, competes for two receptors, TACI and BCMA. APRIL has the ability to stimulate proliferation of various tumor cell lines, including Jurkat T cells and MCF-7 carcinoma cells. Like BAFF, APRIL also stimulates the proliferation of B and T cells. The human APRIL gene codes for at least four alternatively spliced transcriptional variants, which give rise to different isoforms of the APRIL precursor protein. All isoforms can be cleaved by the protease, furin, to release a soluble C-terminal fragment, which comprises the TNF-like receptor binding of the APRIL precursor. It stimulates the proliferation and maturation of megakaryocytes, and promotes increased circulating levels of platelets in vivo. TPO signals through the c-mpl receptor, and acts as an important regulator of circulating platelets. Human and murine TPO exhibit cross-species reactivity. Recombinant Murine TPO is a fully biologically active 174 amino acid polypeptide (18.7 kDa), which contains the erythropoietin-like domain of the full length TPO protein.

The membrane-anchored CD40 Ligand is expressed almost exclusively on activated CD4+ T lymphocytes. Failure to express CD40L leads to "immunodeficiency with hyper-IgM", a disease characterized by failure to produce IgG, IgA and IgE. The soluble form of CD40L is an 18kDa protein comprising the entire TNF homologous region of CD40L and is generated in vivo by an intracellular proteolytic processing of the full length CD40L. Recombinant Murine CD40L is a soluble 16.4 kDa protein containing 149 amino acid residues comprising the receptor binding TNF-like domain of CD40L.
